Both parties named on the check must sign the back of the check. There should be several lines on the back of the check for this purpose.

This depends on how the check is written. If a check is written to someone OR someone then either party can sign as an endorsement. Also if both names are on the account of deposit only one signature is expected by the bank. Otherwise both signatures is recommended.

If you mean that the check is written out to multiple people, both parties must endorse the check. For example, if the check has "Pay to the Order of Richard Jones and Ella Williams," both Richard and Ella must sign. If its just signed by 2 people, endorse it normally.
