Just be real, speak from your heart and you don't have to say a lot, just be authentic.

I always start with, "My name is Inigo Montoya. You killed my father. Prepare to die." Other than recount some endearing or fun time, tell her what she means to you, and say something directly to her wife/husband that welcomes her/him to the family.

First of all don't tell any embarrassing stories or mention past boyfriends. Tell the bride and groom how much you love them both and say something nice about their future lives together. If you have a nice story about their courtship then include this but don't talk too long or drink too much before speaking.

I would mention a few words about your childhood and growing up with her, maybe, tell a funny story about when you were growing up. And plan the toast well so there wont be a dry eye in the house. Also you may want to add a comment on how she wont be able to steal your boyfriends anymore.

Try to be witty, touching and original all at once, which isn't easy any time. Add the stress of the event and even the best-planned salute is difficult to deliver. Say how special your sister is, how clear it is that she and her husband are meant for each other, and how happy you know they will be.
